The cytotoxicity of cytosine hydroxylases and DNA damage potential are the important targets for cancer chemo therapies. In addition to the direct DNA damage caused by apoptosis and necrosis, the interaction of the patient with the tumor is a major factor in the development of metastatic disease. Although the precise mechanisms for the development of metastases in cancers are still poorly understood, it appears that different mechanisms are present for different tumor types and tumor stages. Therefore, many strategies aimed at maximizing and/or minimizing the carcinogenic risk of a given therapy are being developed. These include the development of more targeted therapeutic approaches. This lecture addresses the clinical significance for current anticancer chemotherapy of various aspects of the microenvironment of the tumor and the tumor cells and the cytosine-hydroxylase system.
