Transcription Initiation. The first step in transcription is initiation, when the RNA pol binds to the DNA upstream (5) of the gene at a specialized sequence called a promoter (Figure 2a).
Initiation is the beginning of transcription. It occurs when the enzyme RNA polymerase binds to a region of a gene called the promoter. This signals the DNA to unwind, so the enzyme can ''read'' the bases in one of the DNA strands. The enzyme is now ready to make a strand of mRNA with a complementary sequence of bases.
The goal of transcription is to make an RNA copy of a gene's DNA sequence. For a protein-coding gene, the RNA copy, or transcript, carries the information needed to build a polypeptide (protein or protein subunit). Eukaryotic transcripts need to go through some processing steps before translation into proteins.
Initiation of Transcription in Eukaryotes Unlike the prokaryotic RNA polymerase that can bind to a DNA template on its own, eukaryotes require several other proteins, called transcription factors, to first bind to the promoter region and then help recruit the appropriate polymerase.

Formation of pre-messenger RNA As with DNA replication, partial unwinding of the double helix must occur before transcription can take place, and it is the RNA polymerase enzymes that catalyze this process. Unlike DNA replication, in which both strands are copied, only one strand is transcribed.
The main enzyme involved in transcription is RNA polymerase, which uses a single-stranded DNA template to synthesize a complementary strand of RNA. Specifically, RNA polymerase builds an RNA strand in the 5' to 3' direction, adding each new nucleotide to the 3' end of the strand.
The antisense strand of DNA is read by RNA polymerase from the 3' end to the 5' end during transcription (3' 5'). The complementary RNA is created in the opposite direction, in the 5' 3' direction, matching the sequence of the sense strand except switching uracil for thymine.
