Regulatory sequences are bound tightly and specifically by transcriptional regulators, proteins that can recognize DNA sequences and bind to them. The binding of such proteins to the DNA can regulate transcription by preventing or increasing transcription from a particular promoter.
Gene expression in eukaryotic cells is regulated by repressors as well as by transcriptional activators. Like their prokaryotic counterparts, eukaryotic repressors bind to specific DNA sequences and inhibit transcription. Other repressors compete with activators for binding to specific regulatory sequences.
Translation can be regulated globally (for every mRNA in the cell) through changes in the availability or activity of the helper proteins. For example, in order for translation to begin, a protein called eukaryotic initiation factor-2 (eIF-2) must bind to a part of the ribosome called the small subunit.
However, unlike prokaryotic cells, the eukaryotic RNA polymerase requires other proteins, or transcription factors, to facilitate transcription initiation. Transcription factors are proteins that bind to the promoter sequence and other regulatory sequences to control the transcription of the target gene.
Many transcription factors use one or the other of two opposing mechanisms to regulate transcription: histone acetyltransferase (HAT) activity acetylates histone proteins, which weakens the association of DNA with histones, which make the DNA more accessible to transcription, thereby up-regulating transcription.
Prokaryotic transcription is governed by three main sequence elements: Operators recognize repressor proteins that bind to a stretch of DNA and inhibit the transcription of the gene. Positive control elements that bind to DNA and incite higher levels of transcription.
Bacteria have specific regulatory molecules that control whether a particular gene will be transcribed into mRNA. Often, these molecules act by binding to DNA near the gene and helping or blocking the transcription enzyme, RNA polymerase.
Operon, genetic regulatory system found in bacteria and their viruses in which genes coding for functionally related proteins are clustered along the DNA. This feature allows protein synthesis to be controlled coordinately in response to the needs of the cell.
