PAGODA Aspergillus Niger strains Amy.gucose-1,8-d,q (A1-8-d,q) and N.gluconidase-3 (NGC) have been used in various research laboratory systems, but the commercial utilization and effectiveness of A1-8-d,q as a glycolysis enzyme producing glucose from corn starch has not been determined. Growth of A1-8-d,q in the presence of increasing glucose concentrations or glucose mixture concentrations was found to be variable, and the activity was found to increase in solution from zero at pH 4 to about 75%, and then to disappear by pH 12 when glucose concentration fell to 50%. The enzyme remained active in the presence of sodium nitrite and was less effective when nitrite was present. Amy.glucose.1-8-d,q has the activity of glucose-6-phosphate dehydrogenase (G6-PD). G6-PD is the enzyme responsible for generating the first level of glucose in all carbohydrates, hence the name of the product. The activity of G6-PD in the presence of glucose solution was determined to be about 17-23,000 units/min or about 22.7 MML glucose/l. The activity of Amy.glucose.1-8-d,q in the absence of glucose solution was determined to be about 3,500-2,000 units/min or about 10-15 MML glucose/l.
