What happens if the column runs dry?
If you let the column run dry the silica will start to crack, and you will get poor separation of your compounds. As you run the column, never let the level of solvent go below the level of the silica gel, or you will get poor results.
How do you dry load a column?
Pour dry silica into the pipette by using a small funnel or a paper funnel. Leave about 35 cm of silica-free space at the top. Add solvent to the top of the pipette and allow it to drain. A pipette bulb can be attached to the top and squeezed at this point to speed solvent flow and help the silica pack more densely.
What is a dry load?
Another option is to perform a dry load. With dry loading you blend your dissolved mix with an adsorbent silica, diatomaceous earth, florist, etc. and allow the solvent to evaporate until the mix is a free flowing powder.
How do you speed up column chromatography?
To speed up the process, elute all the solvent using compressed air and allow air to flow through the column for approximately 2 h. This will give dry, free-flowing silica that is easy to pour into the silica waste container.
