Here we discuss the data obtained to date and discuss the implications of such experiments in therapeutics. Meantime works by a mechanism that was not described until recently; its pharmacological properties and cellular mechanisms are very different; however the underlying mechanism appears to have similar mechanisms as those of meantime. Meantime is a selective agonist at the MDA receptor with a k i of 2–5 M and a K d (toxic) of 50–200 km. In vitro studies have shown that meantime affects AD pathology in several cell lines. Recent experiments report a direct effect of meantime on amyloid-beta aggregates formation and also a reduction in the formation of these aggregates as well as a protection against the aggregation of other beta amyloid precursor protein family members on cultured endothelial cells. In vitro studies have also found the ability of meantime to reduce A?1-42 peptide and to reduce the formation of A 1-42 peptide-associated amyloid species, although the clinical impact of these effects were not described. We present preliminary experimental data demonstrating that meantime reduces A?1-42 peptide content in vivo and provide evidence demonstrating a potential therapeutic effect of meantime in amyloid-?-associated neurodegeneration.
