The cellular type has different strains in different parts of the world. The KARA team has isolated the KARA gene, which encodes a transcriptional activator for HSV. A small piece of the KARA gene of three copies, which encodes a protein, was separated from the main KARA protein in a linear array. They found that HSV-1-KARA and HSV-2-KARA were identical. We have named the KARA transcriptional activator the “open reading frame.” The HSV-2 protein is encoded by the HSV-1 gene. We were not able to isolate other genes from the KARA array, but they have been described elsewhere. The KARA protein is contained in an elongated, single-stranded RNA transcript, approximately 1,640 amino acids in length. When HSV binds to a cell wall, the virus induces the closure of the cell wall in a process called phagocytosis, and then the opening of the cell wall. Phagocytosis requires that the cells that are in close contact not release any cytotoxic enzymes. The open reading frame consists of a noncoding RNA gene in the context of another protein that encodes a membrane-anchoring protein. This protein, also known as a plasmid translocator, anchors the open reading frame to the surface of the host cell. This anchoring protein consists of a transmembrane region and an epitome region. The epitome region comprises a small protein domain called an N-terminal domain, and a transmembrane region containing a single amino acid sequence. Phosphorylation by specific protein phosphatase, the transmembrane-bound transmembrane domain, and the epitome regions are responsible for anchoring the open reading frame to the cell. A portion of a sequence in the KARA gene is present in its open reading frame. Our analysis showed that the full open reading frame comprises two large plasmids, which are linked together by the plasmid translocator. The small open reading frame of HSV-1 protein has a transmembrane domain with a transmembrane portion with a domain specific region. The KARA protein encodes the KARA transcriptional activator. Inactivation of the KARA protein results in the closure of the cell wall.
