We developed a highly accurate, stable endoscopic scale system, which has the following features: (1) a small, open-loop system with a small, single injection port that can be rapidly modified to accommodate complex, multi-dose solutions; (2) a user-programmable system that enables the user to fine tune the conditions that must occur before an assay can occur; (3) the ability to process any single, or a combination of multiple, samples without requiring preprocessing; and (4) an automatic calibration with a real-time system or via an automated, self-calibration system via an optical sensor (Fig. 1). A sample was defined as a single, non-migrated, cell, that is, the cell was removed from the liquid to the point where no water was present. The sample was separated from the cell matrix by a non-polarizable poly dextrose membrane. Each sample was subjected to a second, small, open-loop system, which comprises a pump consisting of a sealed, self-contained, pump-injection port that can accommodate multiple samples, to provide sample-to-sample and/or flow-rate-to-sample exchange. Once the sample entered the pump, it was injected with a concentration of a known or prepared reagent and an appropriate volume of fluid is flowed to the sample, in order to form a gel. The pump delivers the fluid to the probe, which is then attached to the sample. The probe was introduced into the system as a fully-assembled sample collection system (Fig. 2A). A second open-loop system, in which sample flow is delivered to the probe through an autotransformable, flexible tubing, was used to assess the efficacy of a series of immunoassay procedures (Fig. 2B–2D). We performed three different tests on the three different types of cells, with different concentrations and mixtures of reagents. Each test requires a few seconds, and a series of independent assays can be performed simultaneously. Fig. 1 Characterization and system design.
