Abstract A multi locus, high-throughput sequencing (MAPS) analysis of bacterial genomes shows the existence of strong homology between antibiotic susceptibility patterns in multidrug-resistant (MDR) and MDR-transmissible (MDR T) strains of Bacillus stearothermophilus (BST). The results suggest that antibiotic-susceptibility genes may have been selected to resist the toxic effect of the LATAM antimicrobial compound (beta-lactam) which caused AIDS. However, the identification of specific ?-lactamases and their phylogenetic position on the bacterial chromosome, may enable identification of the early steps of pathogenesis. © Abstract A MAPS analysis of the microbial genes of Bacillus stearothermophilus is performed to identify MDR and MDR-T strains at specific antibiotic resistance loci. Sequences of these bacterial lineages were compared with previously published sequences and bacterial phylogeny. There is no evidence for the existence of a ?-lactamase. However, there is support for the existence of two genes: an antimycobacterial peptide and a ?-lactamase, in MDR strain. This finding suggests that a ?-lactamase had become evolutionarily important in the life of the bacteria to counteract the toxicity of LATAM. (CC) Abstract This study of the genome of Bacillus stearothermophilus isolates BST431 and BST531 was performed using real-time PCR followed by sequencing analysis of the bacterial genomes. As a result, it was established that the bacterial genome of these two isolates differs at only eight positions. Among them, two genes, which encode antimycobacterial peptides, have been identified. These genes are not found in other antibiotics-resistant species of Bacillus and are similar to some antimycobacterial peptides of Gram-positive bacteria. These proteins may thus serve as molecular switches to prevent the pathogen's attack on the host.
