The study reported here shows that genetic polymorphisms in the cytochrome P450 enzyme system could be used to improve the prediction of toxicity in rats by an integrated system model. The study revealed a more detailed and more accurate prediction of toxicity compared to the previously proposed methods, which used cytochrome P450-based assays alone. More accurate prediction of toxicity was associated with higher cytochrome P450 (CYP) activity in liver biopsies, lower levels of liver transaminase, and lower values for the liver enzyme glutathione reductase. The ability to predict toxicity based on genetic polymorphisms in CYP-450 was superior compared to previous findings. This is important, since genetic polymorphisms that alter the expression of different genes may affect the levels of toxicity. This is important, because some animals may carry a polymorphism in CYP-450 that is more effective in lowering the toxicity of a drug than the genetic polymorphism in the animal model used. However, this is not the case in humans as there is too great a level of variability for such prediction to be accurate.
